We are running several TSC2 data collectors all on 12.49. I picked up one on Ebay for $500 that looks brand new. Seriously it looks better than ours did after one day in the field. I noticed however that it has version 12.10. We are using R8-2's. The basic functionality of it seemed identical from what I could tell. Can I just use this alongside our other units or does it need to be updated? I have no idea what the differences of the various versions are.

Check the release notes on Trimble.com for differences.
You won't be able to copy job files form one to the other, but you can always import points or dc10.70 format files.

I would say that as long as it supports your R8-2s then roll with it - it wouldn't be worth the cost of updating it. Having said that, I would check the software warranty date and see if it's already eligible for any updates. In order to run 12.10, the warranty date has to be at least June 2007. If it was purchased new with 12.10, then the warranty date could be well into 2008, even if it was never updated. If your warranty date is at least October 2007, you're eligible for version 12.22; everything after that, up to and including 12.50, requires a warranty date of October 2008.
